Inflammatory Screening – NF-κB Assay
The nuclear transcription factor NF-κB is a central player in inflammation and immunity. The NF-κB pathway can be activated by pro-inflammatory cytokines such as TNF-α or IL-1β.
Assay Principle
GlycoMar provides a commercial reporter gene assay which uses genetically modified cells (A549 human lung carcinoma cell line) that have been stably transfected with a reporter gene construct expressing secreted alkaline phosphatase (SEAP) under control of NF-κB responsive elements. SEAP is secreted into the supernatant where it is measured using a fluorescent plate reader.
Results
NF-κB activation is calculated as percentage relative to control cells. The assay is suitable for the performance of dose-response studies. Results are reported and summarized graphically, in a format agreed with the client.

Fig 1. Inhibition of NF-κB activity following stimulation with TNF-α% activity relative to controls ± standard error (SEM)

Fig 2. Dose-response study for test agent. % activity relative to controls ± standard error (SEM), IC50 = 0.03 mM
Sample Submission
Samples can be submitted to GlycoMar by prior arrangement, in a number of formats.
Pricing is dependant on sample numbers submitted and turnaround required. Currently the assay is available in a 96-well microplate format offering low – medium throughput. Standard turnaround is 2 weeks from receipt, but faster turnaround is possible on request.
