Lipid Peroxidation and TBARS
(thiobarbituric acid reactive species)
A test for malondialdehyde and 4-hydroxynonenal - downstream lipid peroxidation products primarily found in plasma membranes but also detected in plasma.
Lipid peroxidation is a well-established mechanism of cellular injury, and is used as an indicator of oxidative stress in cells and tissues. Lipid peroxides, derived from polyunsaturated fatty acids, are unstable and decompose to form a complex series of compounds. These include reactive carbonyl compounds, of which the most abundant is malondialdehyde (MDA). Other lipid peroxidation products include 4- hydroxyalkenals such as 4-hydroxynonenal (4-HNE). The TBARS method is commonly used to measure only MDA in biological samples. We have a sensitive assay which measures sum of MDA and 4-HNE. With some modifications it can also be used to measure these products separately.
Assay Principle
This assay is based on the reaction of a chromogenic reagent, N-methyl-2-phenylindole (NMPI), with MDA and 4-hydroxyalkenals. This reaction results in the formation of a stable chromophore which can be measured by a VarioSkan plate reader under specific conditions. The basic principle is the same for MDA, HNE and sum of both but their protocol is slightly different.
Results
The concentrations of MDA and HNE and their sum in unknown samples can be calculated by plotting their standard curves with known concentrations of MDA and HNE.
