Cytokine / Chemokine Screening – Release Assays
An early step in the inflammatory process is activation of inflammatory cells, which results in the release of inflammatory cytokines, such as tumour necrosis factor (TNF)-α and interleukin (IL)-1β. Inflammatory cytokines are central to many of the downstream events, such as leukocyte recruitment, which occur in an inflammatory response.
Assay Principle
GlycoMar’s cytokine/chemokine release assay measures the release of inflammatory cytokines and chemokines from freshly isolated human peripheral blood mononuclear cells (PBMCs) or neutrophils (alternative cell types available on request). The assay can be used to detect both pro-inflammatory and anti-inflammatory activity of test agents. In pro-inflammatory mode the assay looks for the up regulation of key cytokine markers released from cells treated with test agents only. In anti-inflammatory mode the assay uses well characterised inflammatory activators such as PHA, LPS, and IL-1β to stimulate cytokine and chemokine release and looks for down regulation of that release by test agents.
Cytokines and chemokines are determined by individual or multiplex (up to 9 analytes) ELISA; possible analytes include pIL-1β, IL-2, IL-6, IL-8, IL-10, IL-12, IL-13, IFNγ, GM-CSF, TNF-α (full list of analytes available on request).
Results
Results are reported as the amount of analyte released and calculated as percentage release relative to a standard control. Results are reported, summarised graphically, in a format agreed with the client.
