Anti-coagulation Screening – APTT Assay

Polysaccharides such as glycosaminoglycans often possess anti-coagulant activity, the degree of which is dependent on specific structural features. This activity can be of value, as seen with the widely used anti-coagulant heparin and it’s derivatives, but is undesirable when developing polysaccharide-based therapies for other disorders. Determination of anti-coagulant activity of novel polysaccharides is an important part of their evaluation for therapeutic applications.

Assay Principles

The Activated Partial Thromboplastin Time (APTT) assay is an automated functional clotting test based upon the time taken for a plasma sample to clot when activated with a synthetic phospholipid/silica and clotted in the presence of calcium. The APTT assay is sensitive to deficiencies in factors II, V, VIII, IX, X, XI and XII. It is also sensitive to the presence of heparin and non-specific inhibitors.

GlycoMar performs this assay using freshly prepared normal human plasma in an automated (ACL 9000) coagulation analyser. This offers rapid and accurate handling of a high number of samples. The assay is calibrated with International Standard heparin to provide traceable quantitative results.

Results

Anti-coagulation is calculated in International Heparin Units (IU) taken from a standard anti-coagulation curve, prepared using International Standard heparin. Numerical results are reported, in a format agreed with the client. The assay is fully quantitative.

Pricing is dependant on sample numbers submitted and turnaround required. Currently the assay is fully automated offering medium – high throughput.
